Determination of bft gene subtypes in Bacteroides fragilis clinical isolates.

چکیده
A rapid multiplex PCR approach was developed to detect the bft gene subtypes in Bacteroides fragilis clinical isolates. This technique could be used to look at the epidemiology of enterotoxigenic strains of B. fragilis in clinical infections and whether there is a correlation between disease and the presence of B. fragilis enterotoxin.

Identification of a third metalloprotease toxin gene in extraintestinal isolates of Bacteroides fragilis.
To further understand the epidemiology of enterotoxigenic Bacteroides fragilis (ETBF), 89 extraintestinal B. fragilis strains from Seoul, Korea, were examined for secretion of B. fragilis toxin (BFT) by the HT29/C1 biologic assay and for the B. fragilis toxin gene (bft) by colony blot hybridization and PCR. Complete agreement between the three techniques was found. Overall, 34 B. fragilis strai...
متن کاملQuantitative detection of enterotoxigenic Bacteroides fragilis subtypes isolated from children with and without diarrhea.
A rapid real-time PCR (RT-PCR) approach was developed to detect the bft gene subtypes in Bacteroides fragilis isolated from fecal samples. DNA obtained from diarrhea (110) and nondiarrhea (150) samples was evaluated. Subtype 1 was observed in 9 (8.2%) diarrhea and 7 (4.7%) nondiarrhea samples. Subtype 2 was not detected in any DNA samples, and subtype 3 was observed in only 1 diarrhea sample. T...
متن کاملPCR Detection of the Bacteroides fragilis Enterotoxin Gene Relies on Robust Primer Design.
Colonic carriage of enterotoxigenic Bacteroides fragilis (ETBF) is reportedly more common in people presenting with colorectal cancer (CRC) than in healthy controls (1–3), fueling speculation that persistent carriage of these bacteria may “drive” colon carcinogenesis (4). Our aim was to use a bft amplification protocol to determine the prevalence of toxin-producing strains of B. fragilis in CRC...
متن کاملThe C-terminal region of Bacteroides fragilis toxin is essential to its biological activity.
To evaluate the role of the C-terminal region in Bacteroides fragilis toxin (BFT) activity, processing, and secretion, sequential C-terminal truncation and point mutations were created by site-directed mutagenesis. Determination of BFT activity on HT29/C1 cells, cleavage of E-cadherin, and the capacity to induce interleukin-8 secretion by wild-type BFT and C-terminal deletion mutants showed tha...
متن کاملCase-Control Study on the Role of Enterotoxigenic Bacteroides fragilis as a Cause of Diarrhea among Children in Kolkata, India
A total of 874 fecal specimens (446 diarrheal cases and 428 controls) from diarrheal children admitted in the Infectious Diseases Hospital, Kolkata and age and sex matched asymptomatic subjects from an urban community were assessed for the prevalence of enterotoxigenic Bacteroides fragilis (ETBF). Isolates of B. fragilis were tested for the presence of enterotoxin gene (bft) by PCR. The detecti...
